Applications of BBa_K4636000

In this experiment, we performed in vitro transcription of DNA to synthesize RNA and then circularized it. We use RNase R to degrade linear RNA before running the gel.

Each lane sample is as follows:

M: ladder

1: IVT

2: IVT + RNase R

3: CircRNA + RNase R

Lane 1 and 2 are the same IVT RNA sample, both of which used DNase I to degrade template DNA after IVT. RNase R does not have the ability to degrade DNA, but it can degrade linear RNA. Therefore, we can find that the IVT sample without RNase R has a band, but the IVT sample with RNase R has no band on the E-gel EX. The third lane is RNase R, and there are some bands, showing that our sample contains circular RNA.
